Nefericire is built from the negative prefix ne- and fericire, the noun for happiness. Fericire itself comes from the verb a ferici, to make happy, formed on fericit, happy, which traces back to Latin felix, felicem. Adding ne- simply inverts the meaning, giving the standard Romanian word for unhappiness.
