تَغَلَّب comes from the Semitic root غ-ل-ب (gh-l-b), which carries the core sense of overpowering or defeating. The base verb غَلَبَ means simply to defeat or beat someone. Form V, built with the prefix تَ and doubling of the middle root letter, turns this into a reflexive or effortful action: to work at and achieve overcoming something, usually an obstacle, illness, or opponent.
