Cu descends from Latin cum, meaning "with, together with." Latin cum itself traces back to a Proto-Indo-European root meaning "beside, near, by, with," the same root behind the Latin prefix con-/com- seen in words like convene and compose. Romanian kept this preposition short and unchanged, while a separate Latin use of cum as a conjunction developed into the unrelated-looking Romanian word cum, meaning "how."
