"Dest" is a written clipping of "destinatar," the Romanian word for the person a letter or parcel is addressed to. "Destinatar" was borrowed from French "destinataire," which comes from Latin "destinare," meaning to fix or determine. Romanian postal and shipping forms shortened it to "Dest" to pair with "Exp" for "expeditor" (sender). It is a label, not a word people say in conversation.
